NadPrep EZ RNA & DNA Library Co-Preparation Module is specifically developed for mainstream high-throughput sequencing platforms. Featuring a premixed component design, it significantly reduces reagent handling and pipetting steps, enabling simultaneous library preparation of mixed pathogen samples within a single reaction system while remaining fully compatible with liquid-phase hybridization target capture sequencing. This module can be used in combination with the NadPrep Universal Stubby Adapter (UDI) Module and NadPrep Universal Adapter (MDI) Module (for MGI) to support both NovaSeq and DNBSEQ high-throughput sequencing platforms.

Metagenomic libraries were prepared from host-mimicking mixed nucleic acid samples using the NadPrep EZ RNA & DNA Library Co-Preparation Kit. The host-mimicking mixed nucleic acid samples consist of Human Brain Total RNA (Clontech, 636530) and Human Genomic DNA standard (Promega, G1521) mixed at 50 ng:50 ng. Sequencing was performed on NovaSeq 6000 with PE150. The average raw data was 5 M reads pair per sample.

A total of 400 μL of reference standards at different concentration gradients were used for DNA extraction. Pre-libraries were prepared using the NadPrep RNA & DNA Library Co-Preparation Kit (Co-Prep) and NadPrep EZ RNA & DNA Library Co-Preparation Kit (EZ Co-Prep), respectively. 50% of the pre-libraries were performed to hybridization capture using the μCaler HyperMix Hybrid Capture Kit (for Illumina®) and the μCaler RIA Panel v1.0. Sequencing was performed on NovaSeq 6000 with PE150. 0.5 Gb of raw data per sample was used for analysis.

Application Example in Clinical Sample

• Low input amount: If the total nucleic acid input is extremely low (< 1 ng), a reduced library yield is expected and considered normal; subsequent downstream steps can proceed.
• RNase contamination: Use RNase-free consumables throughout the workflow and perform all the operations in RNase-free conditions. RNase contamination will reduce reverse transcription efficiency and low library yield.
• Low sample purity: Residual contaminants in RNA (e.g. chelators, guanidine salts, phenol, ethanol) may inhibit the activity of reverse transcriptase and DNA enzymatic digestion. It is recommended to purify nucleic acid samples according to Step 7: Post-amplification Cleanup using 2X NadPrep SP Beads and elute in Nuclease Free Water.
• Poor sample quality: For low-quality nucleic acid samples, follow the recommendations to increase the no. of PCR cycles appropriately.
